27 November 2020 - RT-PCR Test to Detect SARS-CoV-2 Reveals 10 Major Scientific Flaws at the Molecular and Methodological Level: Consequences for False Positive Results - The published RT-qPCR protocol for detection and diagnostics of 2019-nCoV and the manuscript suffer from numerous technical and scientific errors, including insufficient primer design, a problematic and insufficient RT-qPCR protocol, and the absence of an accurate test validation. Neither the presented test nor the manuscript itself fulfils the requirements for an acceptable scientific publication. Further, serious conflicts of interest of the authors are not mentioned. Finally, the very short timescale between submission and acceptance of the publication (24 hours) signifies that a systematic peer review process was either not performed here, or of problematic poor quality. Eurosurveillance 2020 - External peer review of the RTPCR test to detect SARS-CoV-2 reveals 10 major scientific flaws at the molecular and methodological level: consequences for false positive results. 14 December 2020 - WHO (finally) admits PCR tests create false positives -- The World Health Organization released a guidance memo on December 14th, warning that high cycle thresholds on PCR tests will result in false positives. This new WHO memo states that using a high CT value to test for the presence of Sars-Cov-2 will result in false-positive results. To quote their own words: Users of RT-PCR reagents should read the IFU carefully to determine if manual adjustment of the PCR positivity threshold is necessary to account for any background noise which may lead to a specimen with a high cycle threshold (Ct) value result being interpreted as a positive result. They go on to explain: The design principle of RT-PCR means that for patients with high levels of circulating virus (viral load), relatively few cycles will be needed to detect virus and so the Ct value will be low. Conversely, when specimens return a high Ct value, it means that many cycles were required to detect virus. 16 December 2020 - PCR Test for Coronavirus Questioned by Prominent Scientists - An International Consortium of Scientists in Life Sciences (ICSLS) have submitted a retraction request to the European public health journal Eurosurveillance of a well-cited article published in January 2020,1 2 which contained the first protocol for detecting the SARS-CoV-2 virus using a real-time polymerase chain reaction (PCR) test. The test has been used by the World Health Organization (WHO) as a guideline for other researchers.
